Crucell to move Dukoral® and rCTB bulk production, formulation and fill/finish activities from Sweden to other sites

Leiden, The Netherlands (August 26, 2008) - Dutch biopharma company Crucell N.V. (Euronext, Nasdaq: CRXL; Swiss Exchange: CRX) today announced that it intends to move Dukoral® and rCTB bulk production, formulation and fill/finish activities from Sweden to other sites within the Crucell organization, resulting in additional cost savings beyond 2011.

After thorough analysis and business process redesign, Crucell's operational excellence program, Healthy Ambition, is now being rolled out at full steam. Important elements of the program are: product portfolio optimization, process and infrastructure optimization, network rationalization and further integration and streamlining of various functions. Target savings of €30 million had already been identified and are expected to be achieved by the end of 2009.

Also as part of the Healthy Ambition program, management of Crucell today announced to employees, the intended decision to move Dukoral® and rCTB bulk production, formulation, fill/finish and connected support activities from Sweden to other sites within the organization. The Nordic Sales and Customer Service and Distribution activities remain unaffected. This is expected to result in additional cost savings beyond 2011. Further financial details were not disclosed.

The decision was made with a view to increase the value of the Company by making Crucell operationally excellent resulting in optimized production and financial performance.

The formulation, filling and packaging of Dukoral® in Sweden will stop during 2011 and will move to Spain;
The bulk production of Dukoral® and rCTB will stop in 2012 and will move to Switzerland;
Nordic Sales and Customer Service and Distribution activities will continue to operate from Solna in Sweden;
Research activities will be closed and concentrated in Leiden before year-end 2008.

Outlook 2008:
Crucell expects combined full year 2008 total revenue and total other operating income to grow by 20% in constant currencies [1]. The Company expects higher margins compared to 2007 and positive cash flow

Recombinant cholera toxin B.

rCTB is delivered as a sterile solution in phosphate buffered saline (PBS). No conservation additives are used. The protein concentration usually ranges between 10-15 mg/ml. The rCTB solution is delivered chilled. rCTB is produced in SBL Vaccin’s production facility at Matfors, Sweden, under GMP-conditions.

The molecule

The naturally occurring cholera toxin belongs to the AB class of bacterial toxins. It consists of a pentameric B oligomer that binds to GM-1 receptors (e.g. on the surface of intestinal epithelial cells), and an enzymatically active A subunit that is responsible for the toxicity. The recombinant cholera toxin B-subunit (rCTB) consists only of the non-toxic part of the cholera enterotoxin. The protein is completely free from the toxic A-subunit, which has been genetically deleted from the production strain. rCTB is fermented without antibiotics to ensure safety.

The rCTB molecule consists of five identical monomers with a molecular weight of approximately 11.6 kDa. The monomers are tightly linked into a trypsin-resistant pentameric ring-like structure (see picture) with a molecular weight of approximately 58 kDa.

The protein is included as an active ingredient in Dukoral, the oral cholera vaccine sold by SBL Vaccin.

Dat de research geconcentreerd wordt in Leiden vind ik zeer positief. Onderzoek op één plaats werkt veel efficiënter dan verbrokkeld op meerdere locaties. Unilever is daar met zijn unit in Vlaardingen een goed voorbeeld van. Daar zagen ze in het verleden zelfs kans een zekere mate van synergie te bereiken bij de research in de totaal verschillende sectoren foods en detergents.
De overplaatsing van productie naar Spanje en Zwitserland zal in de beginfase een flinke kostenpost met zich meebrengen. Het is te hopen dat Crucell zich goed laat adviseren door logistieke specialisten.

De verplaatsing van de productie betekent dat een deel van de faciliteiten in Zweden vrij komt voor productie voor derden en ook dat een deel van de GMP faciliteiten voor verkoop in aanmerking komt, mogelijk na het reffen van een voorziening.

Kennelijk kan het verwachte resultaat dit dragen in het 3e kwartaal.

Crucell laat al gedurende lange tijd doorschemeren dat R&D een Leidse aangelegenheid is en zal blijven.
Alle faciliteiten daaromtrendt worden derhalve ook daar gefaciliteerd. Dat er daardoor nu teveel 'scattered' productie capaciteit ontstaat op andere locaties (R&D en productie gaat niet ogenschijnlijk naadloos in elkaar over).
IMHO willen ze sec productie concentreren op locatie (per product/markt) en R&D concentreren in leiden (incl. pre-productie/ productie van klinisch materiaal). Past ook in het plaatje van het valerio gebouw en het huidige afstoten van overbodige productie sites. Just my 2 cents

As the days grow shorter and warnings of frost and the fluffy white stuff make their way into the weather forecast, die-hard sun seekers begin checking for travel deals.
But if the whole idea is to become rejuvenated, then a gurgling, churning stomach that necessitates mad dashes to the washroom can easily wreck a mid-winter vacation.
A little planning and awareness of risk factors can go a long way, says Dr. Mark Wise, who runs The Travel Clinic in Toronto. Research on the Internet or at the library can help, and -especially for more exotic locales -it doesn't hurt to consult a doctor before departure.
He urges people to find out what the risks are at their particular destination.
"It might depend on exactly where they're going or the style in which they're staying or the duration of time that they're going to be there."
"The average trip is a week to Cuba, a week to the Dominican Republic or a week to Mexico and they're all, I'd say, moderately risky," he said.
Twenty to 30 per cent of people going for a week or two will probably get some sort of stomach upset, he said, and a small number of those will get a bad case of diarrhea that totally wipes out their trip.
There's an even higher likelihood of getting sick in places like India, Bangladesh, Pakistan or West Africa, "where the food and water and hygiene is that much worse, poverty is that much worse," said Wise.
Most travellers' diarrhea is caused by the bacteria Enterotoxigenic E. coli, or ETEC.
Dr. Gregory Glenn of Intercell USA Inc., a pharmaceutical company that's working to develop a vaccine patch for travellers' diarrhea, called the ailment a "major disease."
"If you did a bubble of things that travellers come down with, travellers' diarrhea is a giant balloon and everything else is kind of a dot," he said from Gaithersberg, Md.
"It's a major medical need -it's been a very challenging field in which to make a vaccine."
But until medical science comes up with a pill or needle or patch that will fully deliver the goods, Wise urges travellers to learn some rules: "Boil it, bottle it, peel it, cook it, or forget it."
"You just can't go down to these places and drink the tap water and eat under-cooked eggs and maybe the salads that have been washed in water, and expect them all to be clean."
He said he doesn't want to generalize and say that all the fruits and salads in the Caribbean would be a problem every day at every resort.
"But I do say you're only as good as the last person or the last fly that landed on your salad or that touched your food."
Drink bottled water, beer and wine, but be suspicious of the ice cubes, he said.
Make sure that meat is well cooked, he added.
"And I say you don't want to be the last one at the buffet for the lemon meringue pie that's been sitting out for four hours in the sun."
Glenn said people who come down with travellers' diarrhea can end up with post-infectious irritable bowel syndrome or dysfunctional stooling, where it hurts to go.
"Someone will come and say, you know I went to Mexico, I had a big bout of Montezuma's revenge, and it kind of never went away. It lasts for years. So there's a lot of value in preventing the initial event of diarrhea, therefore preventing the irritable bowel syndrome."
In terms of prevention, Wise said an oral cholera vaccine called Dukoral, which costs about $75, will lower the risk of getting travellers' diarrhea by 30 or 40 per cent. The cholera toxin is similar to the toxin produced by ETEC.
The vaccine that Intercell (IOMAI Corp., until a recent merger) is working on contains Escherichia coli toxins. A trial of the patch on healthy adults aged 18 to 64 who travelled to Mexico or Guatemala was reported in the medical journal the Lancet in June, and found 76 per cent protective efficacy against moderate diarrhea, and 84 per cent protective efficacy against severe disease.
A pivotal trial will be held in the summer of 2009 in which researchers hope to confirm the earlier findings, Glenn said. From there, the company would hope to apply for licensure.
But Wise said that no matter how much someone prepares, there will be people who get sick, and they need to know what to do.
"Keep replacing your fluids, which can be done with locally available things like tea with sugar, or soup with salt, or oral rehydration salts," he said.
Travellers may want to pack Pepto- Bismol, an antidiarrheal medication such as Imodium, or have a prescription filled ahead of time in case they need an antibiotic, he said.
"Go prepared, and have fun."

Cholera outbreaks are reported
There have been a number of Cholera outbreaks this year. In September alone there were outbreaks reported from Guinea Bissau, Tanzania, Zimbabwe, Kenya and Iraq. Oral vaccines are available at The Travel Clinic Ltd.

Prevention is always better than cure. There is a cholera vaccine available at The Travel Clinic Ltd, Cambridge and Ipswich, called Dukoral. This is taken by mouth,with two doses given one week apart.
The clinic recommends this to travelers going to at-risk areas who may not be able to be able to source well cooked food or boiled water.

Unicef houdt er rekening mee dat het aantal choleragevallen in Zimbabwe de komende weken toeneemt tot zo'n 60.000. Dat is vier keer zoveel als waar de overheid van Zimbabwe nu van uitgaat, zegt de Unicef-chef in Harare, Monasch.

Volgens officiële cijfers zijn tot nu toe bijna 600 mensen overleden aan de ziekte. Maar Monasch denkt dat het echte cijfer een stuk hoger ligt.

Cholera wordt veroorzaakt door vervuild drinkwater. Door de economische crisis in het land werken de watervoorziening en de gezondheidszorg nauwelijks meer. Van de behandeling van cholerapatiënten komt daardoor weinig terecht.

Bijna 800 doden door cholera in Zimbabwe 10-12-08

Het aantal doden als gevolg van cholera in Zimbabwe loopt snel op.

Gisteren ging de Wereld-gezondheidsorganisatie WHO nog uit van een kleine 600 doden, inmiddels ligt dat aantal op bijna 800. Verder zijn ruim 15.000 Zimbabwanen met de ziekte besmet.

Het is de ergste cholera-epidemie in de geschiedenis van het Afrikaanse land, dat ook nog wordt geteisterd door een politieke en economische crisis.

Opstappen
Westerse leiders, onder wie de Amerikaanse president Bush, hebben president Mugabe opgeroepen op te stappen.

Maar Mugabe wil van geen wijken weten. Volgens hem is de situatie onder controle.

MUSINA - Zuid-Afrika heeft een gebied aan de grens met Zimbabwe donderdag tot rampgebied uitgeroepen als gevolg van de uitbraak van cholera in het buurland.

Het loopt steeds verder uit de hand. Wel vreemd, bij sommige rampen staat heel de wereld op zijn kop om te helpen, en bij andere rampen lijkt het alsof niemand is geinteresseerd. Wordt er binnenkort nog actie ondernomen tegen deze cholera uitbraak?

Zegt alles over hoe selectief de "wereld" is. 1.100 doden pfff. Kijk een tsunami met 240.000 dat gaat ergens over. Zimbabwe, pppf, waar ligt dat dan eigenlijk. En geen olie. Comrade Bob is al 10 jaar bezig het land tot aan de wortels af te breken over de rug van de zimbabwanen die in 1980 na een jaren lange burgeroorlog vrij dachten te zijn. Maar van de regen in drup kwamen aangezien geen omliggend land maar een vinger naar Mugabe durfde uit te steken, bang voor revolte in eigen kring en land. Verandering kan alleen letterlijk over het lijk van Mugabe. De "lobby" om de afbraak van Zimbabwe tot staan te brengen is al zeker 7 jaar in de weer, maar alle deuren bleven dicht. Hoe wrang het ook is, maar misschien is deze epidemie wel de sleutel naar een betere toekomst voor dat indringende land met zachtaardige (hun noodlot!) mensen.
