The present invention relates nucleic acid constructs for the production of recombinant parvoviral (e.g. adeno-associated viral) vectors in insect cells, to insect cells comprising such constructs and to methods wherein the cells are used to produce recombinant parvoviral virions. The insect cells preferably comprise a first nucleotide sequence encoding the parvoviral rep proteins whereby the initiation codon for translation of the parvoviral Rep78 protein is a suboptimal initiation codon that effects partial exon skipping upon expression in insect cells. The insect cell further comprises a second nucleotide sequence comprising at least one parvoviral (AAV) inverted terminal repeat (ITR) nucleotide sequence and a third nucleotide sequence comprising a sequences coding for the parvoviral capsid proteins.

Wat betekent dit voor Uniqure?

Dat is moeilijk te zeggen want daarvoor moet je ook weten wat anderen gepatenteerd hebben rnd dezelfde issue en in hoeverre deze patenten crucial of uniek zijn met betrekking tot het problem dat ze oplossen. In elk geval is uit de inleiding op te maken dat:

Productie van virussen die de dragers zijn van de genen die moeten worden ingebouwd (b.v. AAV5 bij hemofilie B of bij aandoeningen van het Centale ZenuwStelsel)in voldoende hoeveelheden voor therapeutische toepassingen uit zoogdiercellen extreme ineffiecient is en dat dit problem wordt opgelost door uniQures nu gepatenteerde op insectencellen gebasserde technologie. Kortom: voor efficiente productie van je genproduct met je bij QURE zijn!

Voor aandoeningen van het Centrale Zenuwstelsel claimt QURE nu exclusiviteit voor toediening van een AAV5-gebaseerde toediening via een lumbaalpunctie met expressive in zowel de hersenen als het ruggemerg. Hiermee wordt de toediening van AAV5 gebaseerde gentherapieen voor het CZS het unieke domein van uniqure! En in het patent worden echt tientallen aandoeningen genoemd....

Ik denk dat het productiepatent een hele slimme zet is en ook het CZS patent beschermt een platform van waaruit in principe heel veel heel nare CZS aandoeningen kunnen worden bestreden. De waarde van QURe ligt m.i. dan ook in het bieden van platforms, waar een heleboel ontwikkelaars zich beperken tot een paar aandoeningen....

Wat je zou kunnen krijgen is dat partijen die een gen van een bepaalde aandoening in handen hebben zich gaan wenden tot QURE met de vraag of ze er een therapie van kunnen maken en deze kunnen produceren. De regels voor productie zijn streng en de technologie om het gen in een AAV in te bouwen is schaars....

Abstract:

(EN)The present invention relates to the production of adeno-associated viral vectors in insect cells. The insect cells therefore comprise a first nucleotide sequence encoding the adeno-associated virus (AAV) capsid proteins, whereby the initiation codon for translation of the AAV VP1 capsid protein is a non-ATG, suboptimal initiation codon and wherein the coding sequence for one or more amino acid residues have been inserted between the suboptimal translation initiation codon and the codon encoding the amino acid residue that corresponds to the amino acid residue at position 2 of the wild type capsid amino acid sequence of which the first amino acid residue is alanine, glycine, valine, aspartic acid or glutamic acid. The insect cell further comprises a second nucleotide sequence comprising at least one AAV inverted terminal repeat (ITR) nucleotide sequence; a third nucleotide sequence comprising a Rep52 or a Rep40 coding sequence operably linked to expression control sequences for expression in an insect cell; and, a fourth nucleotide sequence comprising a Rep78 or a Rep68 coding sequence operably linked to expression control sequences for expression in an insect cell. The invention further relates to adeno-associated viral vectors with an altered ratio of the viral capsid proteins.
